Abstract

This study was l:onducted to determine the growth induction of inactivated calli and somaclonal variation in barley using 4 two-row and 4 six-row Japanese and Iranian barley selected from cereal collection of the Department of Agronomy, College of Agriculture, University of Tehran. Two weeks after polination, immature embryos were dissected and planted in petri dishes using a soild MS medium with 2
mg/I 2, A-D. Calli were sub-cultured on the same medium after about one-month.
After enough time keeping planting calli in darkness, for growth induction of
inactivated calli different treatments of hormone, light (time, intensity), and cold were
used. Only cold treatment of SOC was suitbale. Cold for IS days and SOC was caused
the most growth induction in inactivated calli. In studying somaclonal variation,
genotypes didn't show significant differences, but with increase of length of planting
period, aneuploidy and euploidy increased.
