The application of four groups of culture media, each with three variations (Tab.1), for embryogenesis and formation of green plants from microspores of F1 hybrid of barley (Hordeum vulgare L.) led to significant differences in plant response, in particular among genotypes. In the first group of media (high levels of Zeatin—riboside and Ficoll) highest frequency of embryo formation from microspores was observed by 1 mg/L cytokinine and 300 g/1 Ficoll with genotype b2 (TR 484/FR 926). Using the second group of media (60 g/l maltose and 300 g/l Ficollor 30 g/l maltose and 400 g/l ficoll) resulted in the highest frequency of green plants with the same genotype. The response of genotypes to third group of culture media (2 mg/L cytokinme, different amount of maltose and sucrose, 200-400 g/l Ficoll) was not favorable and in the fourth group of media (10 g/i starch, 2.5-5.0 mg/L Aminooxyacetic) which is an inhabitor of ethylene production by callus was no plant formation observed.
