Rice is one of the important crops that has a significant role in the country’s economy. Aroma is a trait in rice that highly influences its price as well as its marketing value. In this study efficacy of a RAPD marker linked to the aroma gene namely OPAG-08 was evaluated in F2 segregant generation. During the first year, the cross of Rashti variety to IR28 was obtained, while in the second year, F1 seeds were planted, and then F2 segregant population used for RAPD analysis with OPAG-08 marker. F3 generation seeds were put to allelism test. Phenotypic evaluation indicated that aroma is controlled by two recessive genes and in match with 1:15 ratio (aromatic: non-aromatic) (P=95%). The genotypes of F2 single plants were determined through an analysis of F3 allelism test. F2 genotype and F3 allelism test (phenotype) data were compared, the results indicating that the efficiency of RAPD marker (OPAG-08) for marker assisted selection of aroma genes amounted to 70.62 percent.
